    Backgrounds

    Time-dependent inhibition (TDI) of cytochrome P450 (CYP) 3A4 may result in severe and long-lasting clinical drug-drug interactions (DDIs). However, predictions based on in vitro TDI data tend to over-estimate the risk of DDIs, likely due to conservative inhibitor concentration estimates (Vieira et al, 2014).
