Background: Metabolism and biliary elimination of parent drug are two major routes of drug elimination, affecting drug exposure, possible drug interactions, and adverse events. The primary route of elimination is difficult to predict prior to human dosing, though extensively metabolized drugs are highly permeable in humans.
